measuring of free endotoxin in alum-precipitated vaccine of haemorrhagic septicaemia by limulus amebocyte lysate test
Authors
abstract
haemorrhagic septicaemia (hs) vaccine which is prepared in razi institute is used in endemic areas ofiran. aluminum-hydroxide gel was used as adjuvant for preparing this vaccine. post-vaccinal shock reactionswere the main complaint after use of this vaccine. in a previous study, we could improve the vaccine byalum-precipitation pasteurella multocida cells and removing the liquid phase. in this study, the amount offree endotoxin in aluminum-hydroxide and alum-hs vaccines was determined. it was found that endotoxinlevel was considerably decreased from 0.22 eu/ml to 0.03 eu/ml after alum-precipitation.

full textEndotoxin Contamination of Large Volume Parenterals as Detected by Limulus Amebocyte Lysate as an Alternative to Rabbit Pyrogen Test
The possibility of replacing the rabbit pyrogen test by the Limulus Amebocyte lysate (LAL) test, as a final release test for Large Volume Parenterals (LVPs) was investigated. The sensitivity and specificity of the LAL test and rabbit pyrogen assay were studied by means of artificially contaminated parenterals. Various LVPs (mostly containing electrolytes) were spiked with 0.5 and 5 EUlml o...
full textSingle-step, chromogenic Limulus amebocyte lysate assay for endotoxin.
A new reagent for the chromogenic Limulus amebocyte lysate (LAL) assay is described. LAL was formulated for optimal performance in either an endpoint procedure or a kinetic procedure with the chromogenic substrate, buffer, and LAL components colyophilized as a single reagent. The kinetic chromogenic method required an incubating microplate reader coupled to a computer for collection and analysi...
full textBiomedical Applications of Limulus Amebocyte Lysate
This year celebrates the 30th anniversary of the licensing of Limulus amebocyte lysate (LAL) by the US Food and Drug Administration (FDA) as a test for the presence of endotoxin in biologicals, pharmaceutical drugs, and medical devices. LAL is currently recognized by several major pharmacopoeias and is used worldwide. That a suitable alternative for the detection of endotoxin has not supplanted...
full textEndotoxins and Their Detection with the Limulus Amebocyte Lysate Test
In contrast to the detailed descriptions of techniques provided by Larsson, a chapter on the measurement and analysis of acetylcholine written by Israel Hanin takes the form of a brief but comprehensive survey of all of the methods for the quantitation of acetylcholine introduced between 1933 and 1981. Each method presented is referenced and briefly critiqued. This is followed by an article by ...
full textComparison of the Limulus Amebocyte Lysate Test and Gas Chromatography-Mass Spectrometry for Measuring Lipopolysaccharides
mass spectrometry analysis of lipopolysaccharide-derived 3-hydroxy fatty acids. Gram-negative cell walls were also measured by using two-dimensional gas chromatography/electron-capture analysis of diaminopimelic acid originating from the peptidoglycan. The correlation between the results of the Limulus assay and those of gas chromatography-mass spectrometry for determination of the lipopolysacc...
